In vitro propagation of Platicerium andinum Baker from spores

Abstract

The forests of San Martin Region, habitat of Platycerium andinum B. have been destroyed in an excessive way, caused by anthropogenic activities, which has led to the loss of the species in its natural habitat, adding to it the extraction for its exuberant beauty to be market as an ornamental plant, its spores are also difficult to germinate under natural conditions, in addition, there is no methodology for the in vitro propagation of this species. The objective of this research is to determine the adequate concentration of sodium hypochlorite to obtain pathogen-free P. andinum spores and to evaluate three farming media to determine the most suitable medium for the propagation of gametophytes. The spores were obtained from the fertile fronds of adult plants by scraping them off. The germination of the spores was evaluated after 10 days, time in which they began to germinate and after 30 days there was already abundant gametophyte tissue; it was evaluated through the spore germination rate (GI) using the Braun-Blanquet abundance-coverage scale, reaching scale 5 after 60 days (any number of gametophytes with coverage greater than 75%). Regarding the determination of the best farming method for the in vitro propagation of gametophytes, three MSB media (T1, T2 and T3) were used with additives of 0.4 ml. of thiamine, 0.5 of nicotinic acid, 2 grams of activated carbon and 20 grams of sucrose; with 100 ml of coconut water in T2, and 200 ml in T3, obtaining the best result for treatment T1: (M and S Basal, with the addition of 0.4 ml. of thiamine, 0.5 of nicotinic acid, 2 grams of activated carbon and 20 grams of sucrose).
